What is Melanotan I?
Melanotan I (afamelanotide) is a synthetic analog of alpha-MSH with two amino acid changes that make it more stable. It stimulates melanin production.
An implant form, Scenesse, is approved for erythropoietic protoporphyria.
How Melanotan I acts in the lab
Melanotan 1 acts on MC1R, the melanocortin receptor on pigment cells. Activating MC1R switches melanocytes from making pale pheomelanin to darker eumelanin.
Two swaps in the alpha-MSH sequence (norleucine at position 4 and D-phenylalanine at position 7) make it far more potent and longer lasting than the natural hormone. It’s more selective for MC1R than Melanotan 2.

Melanotan I 10mg reconstitution and concentration
Concentration is just the amount in the vial divided by the liquid you add. With 10mg in this vial:
- 1mL of diluent gives 10mg/mL
- 2mL of diluent gives 5mg/mL
- 3mL of diluent gives 3.33mg/mL
Go with whichever number is easiest to work with in your protocol, and label the vial with the concentration and the date you mixed it.
Research diluent solution or bacteriostatic water both work. Add it slowly so it runs down the side of the vial, then swirl or roll until everything dissolves. Shaking can shear peptide chains. Skip it.
Storing Melanotan I 10mg
Unopened, keep Melanotan I 10mg in a fridge at 2 to 8°C, or in a freezer at -20°C if it’ll be a while before you use it. Keep it out of direct light. Once mixed, keep it refrigerated and use it within about four weeks.
